City types in Spore differentiate each other only by their architectural design.
[edit] Regular City
In board terms, the definition of a regular city is one that exists either on land without the aid of a protective dome and is built from raw materials, such as clay or steel. This type of city will probably be the most common of all types due to it being the most recognizable form of a city, as well as the easiest to make.
[edit] Domed City
Domed cities are collections of buildings placed under a protective barrier to allow its inhabitants to survive the elements in comfort. This can range from an underwater city built on land, a land based city build underwater, or either built in a harsh environment. Created automatically using a similar architecture of your main city, these colonies slowly gain resources for your civilization. If the environment around the dome is suitable for the species to survive without it, the dome becomes unnecessary and is removed.
